Improved Tissue Distribution, Renal Clearance and Toxicity Profile of Functionalized Carbon Nanotubes

چکیده
In this communication we discuss the in vivo parameters such tissue distribution, body clearance and toxicity following administration of carbon nanotubes (CNT). For this propose, therapeutically-relevant doses of serum protein-coated purified CNT (pCNT) and functionalized CNT (f-CNT) were administered intravenously in rodent species. Striking differences were found between the liver and lung tissues of animals injected with nanotubes. pCNT exhibited significant accumulation in both tissues 24 h post-administration. f-CNT circulated in systemic blood circulation for 5 min, accumulated in the renal cortex and rapidly trespassed the glomerular filtering system and excreted in urine in the absence of any tissue accumulation or damage. The biocompatibility and biodistribution of CNT appears to be dramatically dependent on the degree of surface functionalization of these nanostructures, making fCNT viable material for a variety of biomedical applications.

Getting rid of the tubes: An assessment of the retention of functionalized multi-walled carbon nanotubes (MWNTs) in the organs of mice was carried out using single photon emission computed tomography and quantitative scintigraphy (see scheme). Increasing the degree of functionalization on MWNTs enhanced renal clearance, while lower functionalization promoted reticuloendethelial system accumulat...

Carbon nanotubes (CNT) are intensively being developed for biomedical applications including drug and gene delivery. Although all possible clinical applications will require compatibility of CNT with the biological milieu, their in vivo capabilities and limitations have not yet been explored. In this work, water-soluble, single-walled CNT (SWNT) have been functionalized with the chelating molec...

This paper investigates the effect of different methods of carbon nanotubes distribution in a thin matrix on static and dynamic behavior of the nanocomposite. Five different symmetric patterns of distribution are considered, including four parabolic patterns and a linear one. For each pattern, the effective mechanical properties of the resultant nanocomposite are calculated using the rule of mi...

Carbon nanotubes are promising new materials for molecular delivery in biological systems. The long-term fate of nanotubes intravenously injected into animals in vivo is currently unknown, an issue critical to potential clinical applications of these materials. Here, using the intrinsic Raman spectroscopic signatures of single-walled carbon nanotubes (SWNTs), we measured the blood circulation o...
